Djokovic was denied his landmark title back in March, when he went down to Jakub Mensik in the final of the Miami Open.
And despite falling a set behind against Hubert Hurkacz on Sunday, the 24-time grand slam champion hit back to prevail 5-7 7-6 (7-2) 7-6 (7-2) in three hours and eight minutes.
Djokovic went into the ATP 250 in poor form, having lost his first match in four of his last five tournaments.
But the Serbian clicked through the gears through the week, and has now joined Jimmy Connors and Roger Federer on the list of men with 100 ATP titles under their belt.
Now, Djokovic's attention will turn to Roland-Garros, as he hunts a record-breaking 25th major title.

Djokovic has now won at least one ATP-level event for a 20th consecutive season, becoming the first player in the Open Era to achieve the feat.
His most recent victory had come back at last year's Olympics, on the same clay courts where the Serbian will hope to shine over the next two weeks.
With 72 such wins to his name, Djokovic has claimed the most big titles - grand slam, ATP Masters 1000, ATP Finals and Olympics - of any player since the inaugural ATP Tour season in 1990.
